The
specificity of IQ2000 IHHNV nested Detection and Prevention System has
been
proved.
1. This
kit can specifically detect the
IHHNV isolated from different geographic area.
2.
This
kit can specifically pick up the
infectious IHHNV without interacting with the integrated IHHNV in the
P. monodon.
Dr.
Kathy Tang-Nelson from the University of Arizona tested IQ2000 IHHNV
nested Detection
and Prevention System (a nested PCR with sensitivity of 10
copies/reaction ) on
the specificity of this kit with samples of (1) IHHNV isolates, (2) P.
monodon DNA containing IHHNV-related
sequences, and (3) DNA extracted from SPF penaeids and aquatic
organisms that
were used as feed for shrimp culture.
The
result showed that there were no non-specific reactions with 13 samples
of SPF
penaeids, lobster, crabs, artemia, krill, polychaetes. This PCR kit did
not
react to the 42 samples P. monodon
containing integrated IHHNV-related sequences that were collected from
Tanzania, Australia,
shrimp farms in Malaysia,
and India.
Also, this kit is specific to IHHNV and reacts to purified IHHN virions
and
IHHNV isolates from America
(Hawaii, Brazil,
Belize, Mexico),
Oceania, and SE Asia.
There is no cross
reaction to another shrimp parvo-like virus, HPV.
Users
who concern about integrated IHHNV and need higher analytical
sensitivity can
choose this kit to fit for their detection purposes. However, IQ2000
IHHNV
Detection and Prevention System (one step PCR with sensitivity of 100
copies/reaction) is still available for users who only detect IHHNV on
penaeid
shrimp other than P.
monodon.
